3. Deposition of the Raw Sample
a) Spot Mode:Using capillarity, either via an HPLC syringe or a capillary, the solute penetrates the adsorbent layer to varying degrees. Deposits made with a syringe tend to be finer compared to those made with a capillary. When using this method, the spots might appear slightly distorted. Load the smallest amount possible to avoid overloading-induced dispersion, which can impact compound resolution. Avoid touching the TLC plate with the depositing tool to reduce excessive penetration of the solute into the plate's layers. Spot density and dispersion may change depending on the solvent polarity. b) Spray Mode:With a spray bottle, ensure the spray rate is slower than the solvent's evaporation rate to avoid splashing. This method results in minimal solute penetration into the adsorbent layer, keeping the solutes largely on the surface. Solvent of DilutionChoose a solvent with low elution power, ensuring it is highly pure. It should be polar enough to dissolve the sample but not excessively so that it evaporates too quickly. Limit the use of bases and acids. Avoid viscous or high boiling point solvents like N,N-Dimethylformamide, DMSO, BuOH, or water, as these can slow down the migration of the solvent. Always allow the plate to dry thoroughly between successive deposits.Solvent of ElutionAdjust the elution strength based on the polarity of the compounds to maintain Rf values within the optimal range (0.15 – 0.35). Note that the mobile phase velocity isn't consistent across the entire plate. Use the same mobile phase for both TLC and flash purification. For a seamless transition to Flash purification: - Measure solvents precisely using separate flasks (double-check the flask's accuracy). - Use a syringe for low-volume measurements to enhance precision. |
