Labeled streptavidin-biotin method (LSAB)

The labeled streptavidin-biotin method (LSAB method) is a labeled avidin technology that was introduced in the 1980s.

Basic reagents:
1 primary antibody;
2 biotinylated secondary antibody;
3 Horseradish peroxidase-labeled streptavidin.

experimental method:
1. Rinse the fixed specimen with PBS;
2. Soak the smear with 1% H2O2 or 1% H2O2 methanol for 10-20 min at room temperature to seal the endogenous peroxidase;
3. TBS rinses 3 times, each time 1 min;
4. Incubate with normal serum of secondary antibody for 20 min at room temperature;
5. Add the primary antibody and leave it at room temperature for 1 hour or 4 ° C in the refrigerator overnight;
6. TBS rinses 3 times, each time 1min;
7. Drop the secondary antibody and leave it at room temperature for 30 min;
8. Incubate the specimen with the biotinylated secondary antibody for 10 min at room temperature;
9. TBS rinse 3 times, each time 1min;
10. Incubate with diluted LSAB for 10 min at room temperature;
11. Repeat step 9;
12. Develop color with DAB or AEC;
13. Rinse repeatedly with distilled water;
14. Counterstaining with hematoxylin;
15. Covering;

ETR oxygen cylinder filling plant is consisted of Atlas Copco air compressor, Parker dryer and filters, air buffer tank, ETR Psa Oxygen Generator, oxygen buffer tank, oxygen receiver tank, diaphragm oxygen booster, filling station and control cabinet.

Compressed air is purified through the air dryer and filters to a certain level for main generator to work with. Air buffer is incorporated for smooth supply of compressed air thus to reduce fluctuation of compressed air source. The generator produces oxygen with PSA (pressure swing adsorption) technology, which is a time proven oxygen generation method. Oxygen of desired purity at 93%±2% is delivered to oxygen buffer tank for smooth supply of product gas. Oxygen in buffer tank is maintained at 4bar pressure, then reach to 150bar to filling cylinders with an oxygen booster.
